What are Dark Spots?

How are Dark Spots formed?


Dark spots are clinically known as hyperpigmentation, which is specifically caused due to abnormal stimulation of melanogenesis in different localized regions of the skin. According to D’Mello et al. (2016), melanogenesis is a process that helps the skin in getting its color as melanocytes start creating melanin. 


It has been noted that in normal physiological conditions the melanocytes, which are located at the basal layer of the skin, synthesize and distribute the pigment melanin uniformly throughout the skin, which helps in delivering the skin its consistent and uniform tone.

However, whenever the skin is exposed to certain triggers like UV exposure etc, the melanocyte activity gets disbalanced, resulting in increased production of melanin in specific areas. This increased rate of melanin production is a part of the natural defense mechanism of the skin, keeping it safe from the external aggressors (Hong, Lim and Kim, 2026). This increased rate of melanin production is often not distributed uniformly throughout the skin regions, thereby producing an uneven appearance. 


Moreover, Hurbain et al. (2018) found that the uneven distribution of concentrated pigment creates keratinocytes with the help of specialized organelles, known as melanosomes. In the keratinocytes, this excess pigment starts accumulating in higher concentrations (Hasegawa et al., 2015). With time, the melanosome-infused keratinocytes shift towards the skin’s surface as a part of the epidermal turnover process, which enables the accumulated pigment to become visible as dark spots.

What is Kojic Acid?


Kojic acid is an organic compound, made with a wide variety of fungal species like Aspergillus oryzae etc, while being classified as a heterocyclic compound (Ola et al., 2019). It was observed that kojic acid is a colorless and needle-like crystal that is often soluble in different liquids like water, ethanol, and ethyl acetate (Nih.gov, 2026). 


While considering the function of Kojic acid, it was noted by Mohamad et al. (2010) that it is a weak acid as it has got a hydroxyl group at the carbon-5 position. Thus, being a weak acid, it easily forms metal salts after coming into contact with metals like zinc copper, nickel, etc. 


Therefore, the reactive multifunctional structure of the compound has made it popular in a diverse range of industries, starting from food processing, healthcare and cosmetic sectors. How Kojic Acid Works on Dark Spots?

  • Controls Melanin Production: Kojic acid is primarily used by most skincare and cosmetic sectors due to its capability of inhibiting tyrosinase. It was observed that melanocytes, the skin cells, create melanin with the help of an enzyme called tyrosinase. Mucha, Skrzydlewska and Gęgotek (2025) stated that the skin cells create melanin as the body’s natural defence mechanism against external stressors like UV, pollution, etc. Tyrosinase works in two critical steps while helping melanocytes to create the required melanin content. Initially tyrosinase changes an amino acid called tyrosine into L-DOPA while later it changes L-DOPA into dopaquinone, which results to the creation of melanin (Slominski, Zmijewski and Pawelek, 2011). Without tyrosinase this pigment creation chain would cease to exist. Here’s where Kojic acid steps in. Application of Kojic acid on the skin surface helps in binding with the copper ion present in tyrosinase while disabling its catalytic activity to complete the melanin synthesis, thereby preventing hyperpigmentation, dark spots and post-acne marks.

  • Provides Antioxidant Protection: Application of kojic acid to skin helps in gaining its antioxidant protection against free radicals, which are mostly generated through environmental stressor like UV radiation, pollution, etc. According to Eskandani et al., (2010), oxidative stress often triggers the tyrosinase activity while causing an increased production of melanin. In this connection, use of kojic acid helps in extending antioxidant protection while preventing oxidative stress and protecting against free radicals, thereby reducing the overdrive of tyrosinase activity and controlling melanin production (Saeedi, Eslamifar and Khezri, 2019).   • Manipulates Cellular Signaling: Kojic acid influences the keratinocytes, which are the outermost skin cells that surround the melanocytes, to secrete interleukin-6 (IL-6). Fu et al. (2020) stated that interleukin-6 (IL-6) is a cytokine that blocks the melanocytes from receiving signals on different external stressors, thereby inhibiting tyrosinase to produce melanin. This activity helps indirectly in regulating the melanin production by interfering with the cell-to-cell communication chains, resulting in a fair and bright skin complexion.

  • Antimicrobial and Antifungal: With antibacterial and antifungal properties, kojic acid helps in disrupting the cell membranes and different metabolic processes of the microorganisms that grow on the skin surface (Zilles et al., 2022).
